结合研究目标构建模型，并不是药物所有的 ADME 机制都需要整合到模型中，有时简单、可靠的模型就能解决问题。
模型存在假设是正常现象，但需要进行合理解释或敏感性分析。
预测出现偏差，除了建模参数不全或数据质量问题之外，还有可能是没有充分考虑药物的 ADME 机制，因此优化模型的过程实际也是在探讨可能的机制。
模型优化并不是任意的修改模型参数，而是需要结合已有的数据、个人经验等合理的优化模型参数，而通过个人经验优化的模型参数需要有理论支持或进行数据验证。
PBPK 建模过程属于“Bottom-up”，但往往需要结合“Middle-out”优化模型，甚至可能用到“Top-down”分析建立的模型。
